Abstract: Immunoassays are methods that use antibodies to measure extremely low concentrations of compounds in nanogram (1 ng=10-9 g), picogram (1 pg=10-12 g) and even femptogram (1 fg=10-15 g) levels that can be measured in a gram or less of biological sample. A wide range of compounds can be measured by this technique as pesticides, proteins, proteic hormones, steroids, drugs, viral or microbial antigens. ELISA (enzyme linked immunosorbent assay) is a technique based on the ability of non-labeled antigen (e.g. pesticide) in a specific volume of standard solution or in an unknown sample to compete with a fixed of amount of enzymatic labeled antigen for a limited number of binding sites of a specific binding antibody protein. Project has as objective development of ELISA technique for detection of pesticide contaminant 3,6 dichloro-2-methoxybenzoic acid from alimentary and environmental samples. 3,6 dichloro-2-methoxybenzoic acid is one of the herbicide most used in agriculture to control and destroy of the weeds that can affect agricultural crops. The remanence of this organochlorurate compound in alimentary products, transfer and contamination of ground water in the areas where this pesticide is used require the monitoring of this chemical in order to establish the contamination level of the alimentary products and the environmental factors (water, soil). Having a great sensitivity, the ELISA technique can be used in assays of this pesticide in the alimentary and environmental samples. The objective of the project is obtaining of the specific reagents used in ELISA technique: immunogenic conjugates pesticide-protein (used for the obtaining of antipesticide antibodies) antipesticide antibodies, enzymatic markers, standards and other reagents necessary for ELISA technique.

1. Obtainment of immunogenic conjugate 3,6 dichloro-2-methoxybenzoic-protein and physical-chemically characterization (2007-12-10)
2. Obtainment of anti 3,6 dichloro-2-methoxybenzoic-antibodies. Immunization. Obtaining of antisera, immunological characteristics of antisera, titre, affinity against antigen (2008-06-15)
3. Obtainment of enzymatic markers: 3,6 dichloro-2-methoxybenzoic-peroxidase, 3,6 dichloro-2-methoxybenzoic-alcaline phosphatase, anti 3,6 dichloro-2-methoxybenzoic antibody-peroxidase, anti 3,6 dichloro-2-methoxybenzoic antibody-alcaline phosphatase. 4. Kinetics studies of the antibody-antigen binary system antibody antipesticide-pesticide (2009-12-15)
